Safety evaluation using intratracheal instillation method

Today, Intratracheal instillation method is used to evaluate the lung injury of substances, nanomaterial, etc. exposed through the respiratory tract. This method is also useful for screening local pulmonary toxicity of drug candidates. Moreover, verification for the alternative to inhalation exposure is progressing.
This method is easy and inexpensive, and has advantage for clarifying dose-response relationship being able to estimate exposure level, and is suitable to solve mechanisms of the biological responses.
This method is available as a screening for primary candidate substances to explore inhalation toxicity.
Repeated intratracheal instillation to rats or mice is achievable by our experts.

Male and female F344 rats, 10 weeks of age are used.

After 13 weeks from the commencement of treatment, organ weight, BALF inspection, histopathology for lung inflammation reaction are performed.

After 104 weeks from the commencement of treatment, carcinogenicity are estimated.

Eight weekly instillations are enough to evaluate carcinogenicity compared with vehicle control.

It is useful as a screening method to evaluate the lung injury by aspiration.

Intratracheal instillation method is rational method rather than intraperitoneal administration

for extrapolation.
